This is probably the oddest question ever conceived for a topic.
How about this. Close the brothel and sell the building it was operated in. Use the money to get the ladies tested/treated for STDs.

Well, opening a brothel in the red light district is like impossible, 99% of all the brothels, perhaps even 100% is run by criminals by the dutch mob. You will not get easely in that world. An old coleage of me ran a escort service wich did pretty good, 150 euro/hour 6 girls working 5 hours a day. 750*6 = 4500 a day obviously not paying tax since he imported the hookers from bulgaria. Than later he got in trouble with those bulgariun dudes, now he is missing lol. too bad i cant drive his new mercedes e330 anymore.

I’d fix it up.
Turn it into a real classy joint. Blues in the lobby.
Redo the walls and shit. Make the lobby look erotic.
I’d make it so it’ll turn to a social place where everyone can hangout and shit and the whores will be just a way to end the evening. 
Have a burlesques show on every night.
An open bar with limit of three drinks per customer.

Thats the spirit .. would the girls live in or have places outside & just work the joint ? If so would you charge them for rooms or take half what they make of em ?

Nah.
I'd let them live here as long as they keep their rooms clean.
So customers will want to come back.  Sanitation is pwn! 
If they can't keep their rooms clean I just won’t let them do anything until the room is spotless.
I wouldn't charge them much. Just enough to keep the place running. Like 20% from every girl working there.
If they don't want to live here, I'll charge them a room rental fee of 40% of what they'll make.
The non working girls will work as waitresses in the bar downstairs. Take care of soon-to-be customers. Clean up. Keep company and just make the place pleasant and returnable.
